Betont hellfarbig wird hier die biblische Szene der Kreuzabnahme gezeigt. In den Lokalfarben jedoch spricht deutlich der Manierismus, wobei hier die nahezu selbstleuchtenden Rosa- und Gelbfarben komplementär zum Grün in den Gewändern zum Ausdruck kommen. Ausgehend von der Schule des Florentiners Jacopo da Pontormo hat diese Farbgebung auch in die Renaissancemalerei Tirols und nördlich der Alpen Eingang gefunden. Bemerkenswert ist hier auch die in helle Rosafarbe getauchte Architektur der Stadt Jerusalem oben links
Die Darstellung zeigt sämtliche im Bibeltext genannte Figuren beim Geschehen der Kreuzabnahme. Textgemäß erfolgte dies noch bei Tageslicht, was der Maler ganz offensichtlich zeigen wollte, ganz im Gegensatz zu vielen gleichthematischen Werken anderer Künstler der Zeit. Die einzig dunklen Aspekte im Bild finden sich im Mantel der Maria, die unterhalb des Kreuzes niedersinkt, sowie in der Grabeshöhle rechts am Bildrand, in der die nachfolgende Grablege synchronoptisch gezeigt wird. Die Leidenswerkzeuge – Kreuznägel mit Hammer, Essigschale mit Schwamm-, sind hier am Boden im Vordergrund ins Bild gebracht, die Dornenkrone jedoch wird von einem der Männer auf der Leiter gezeigt – was als ein außergewöhnliches Motiv zu sehen ist.
